Using RT-PCR, we measured the ratio of HN to L mRNA accumulated in cells infected with wt and rSV5s containing altered gene تعمیرات جاروبرقی در تهران junction sequences. The results (Fig. 5A) showed that while substitution of the gene junction sequences significantly changed the relative amount of HN and L mRNA, there was no change in attenuation level. This result suggests that the SV5 GE, IG, and GS sequences are not essential for attenuation of wt viruses and that the relative position of the gene start site within a hexamer is important.
Using a similar approach, we investigated whether the SV5 gene junction sequences influence gene expression in CV-1 cells infected with recombinant virus genomes encoding GFP between the HN and L genes (12a). RNA probes annealed to HN and L mRNAs in vRNA-containing poly(A) chains were used to detect mRNAs in cellular fractions that had been treated with RPA. This revealed that a substitution of the NP-V/P or HN-L gene junction sequences did not significantly alter the amount of mRNA accumulated in these cells.
